5-Amino-1MQ (NNMT Inhibitor)

A cell-permeable quinolinium compound, CAS 42464-96-0. It sits in this catalogue because that is where buyers look for it — but it is not a peptide, and it should not be characterised like one.

Pricing and pack sizes

SizeCat. no.PackPrice per kitPer vial

10 mg10AM10 vials$58.00$5.80
50 mg50AM10 vials$88.00$8.80

Prices are in USD and are per kit of 10 vials — not per vial. Both figures are shown so the two can be compared directly; a "per vial" price quoted without saying so is how the same material ends up looking ten times cheaper than it is.

Larger quantities, mixed kits and custom synthesis are quoted on request. Current lot documentation accompanies every quotation.

It is not a peptide — and that matters analytically

5-Amino-1MQ is a methylquinolinium salt: a fused bicyclic aromatic cation with a primary amine, molecular weight 194.66 as the hydrochloride. There is no amide backbone, no residue sequence, and nothing for a peptide-sequencing workflow to do.

Knowing this changes three practical things:

  • Detection wavelength. The quinolinium chromophore absorbs strongly in the UV, so purity is monitored at 254 nm rather than the 220 nm used for peptides. A COA that reports "HPLC 220 nm" for this compound is copying a peptide template.
  • Identity. ESI-MS gives the molecular ion, but for a small molecule that is weaker evidence than it is for a peptide — isomers and positional analogues share the same mass. 1H NMR is the appropriate confirming technique, and is available on request.
  • Salt form. The hydrochloride is hygroscopic and the stated potency depends on the salt factor. A purity figure quoted without salt form is not a potency figure.

We list it under peptides because that is where the demand is, not because the chemistry belongs there. If a supplier describes this compound as a "peptide" and offers a peptide-style COA, that tells you how the rest of their documentation was produced.

Research context

The following summarises mechanisms reported in preclinical and in-vitro literature. It is provided for scientific orientation only and does not describe any approved or established clinical application.

NNMT inhibition

Nicotinamide N-methyltransferase transfers a methyl group from S-adenosylmethionine to nicotinamide. Inhibiting it spares the methyl donor and shifts the SAM/SAH ratio, which in turn influences methylation capacity and the availability of nicotinamide for NAD+ salvage. 5-Amino-1MQ is used as a tool compound for that axis.

Adipocyte and metabolic models

The most-cited cell work uses adipocyte and hepatocyte lines, reporting reduced lipid accumulation and changes in energy-metabolism gene expression. These are cell-culture observations at micromolar concentrations.

Note on evidence status

The literature is early-stage: cell models and a limited number of rodent studies. There is no clinical trial data, and the compound is not an approved medicine anywhere. Its usefulness in this catalogue is as a tool compound, not as a candidate therapeutic.

Analytical verification

  1. Purity — RP-HPLC. C18 column, gradient acetonitrile/water with 0.1% TFA, UV detection at 254 nm, matched to the quinolinium chromophore rather than the 220 nm peptide default.
  2. Identity — ESI-MS. Molecular ion at m/z 159.1 [M−Cl]+ for the cation, consistent with C10H11N2+.
  3. Salt and water content. Reported on the COA, because the hydrochloride is hygroscopic and potency is salt-dependent.

Look up your lot in the COA registry for the chromatogram and mass spectrum of the specific batch.

Preparing a standard solution

For preparing calibration standards for instrumental analysis. Not a preparation guide for any biological or human application.

  1. Weigh on a calibrated analytical balance; the compound is crystalline and transfers cleanly.
  2. Dissolve in water for aqueous methods, or DMSO for stock solutions where aqueous solubility limits the working concentration.
  3. Filter 0.22 µm before injection if any undissolved material remains.
  4. Protect from light — the quinolinium ring is photolabile over extended exposure.
  5. Store stock solutions at −20 °C, protected from light, and avoid repeated freeze–thaw.

Frequently asked questions

Is 5-Amino-1MQ a peptide?

No. It is a small-molecule methylquinolinium salt, molecular weight 194.66 as the hydrochloride, with no amide backbone and no residue sequence. It appears in peptide catalogues because that is where buyers search for it.

What is the CAS number?

42464-96-0.

Why is purity measured at 254 nm instead of 220 nm?

The quinolinium chromophore absorbs strongly in the UV around 254 nm. The 220 nm wavelength used for peptides is the wrong detection point for this molecule — a COA that reports 220 nm for 5-Amino-1MQ is using a peptide template.

How is identity confirmed?

ESI-MS gives the molecular ion; for a small molecule of this size, NMR is the stronger confirmation because positional isomers share the same mass. 1H NMR data are available on request.

What salt form do you supply?

The hydrochloride. Salt factor and water content are reported on the COA because the material is hygroscopic and potency depends on them.

What pack sizes are available?

5 mg, 10 mg and 50 mg vials, supplied as 10 vials per kit. Prices per kit are listed on this page; larger or mixed orders are quoted on request.

How should it be stored?

−20 °C, desiccated, protected from light. The quinolinium ring is photolabile, so amber glass or a dark box is preferred for long-term storage.

Is 5-Amino-1MQ approved for human use?

No. It is not approved as a medicine, food, cosmetic or veterinary product in any jurisdiction, and is supplied strictly as a reference standard for in-vitro laboratory research.
